It's technically possible, but not practical. It's the exact same thing as a down stroke, except instead of using your thumb to mute, you can use your index finger I guess. All pinch harmonics is, is touching the string right after you pick it. It wouldn't make a difference if your coming up or going down, just as long as you touch the string after you play it.

or if your really good you can pluck the string with your thumb and then hit it with the pick having the same effect and a normal pinched harmonic

if you hold the pick with your thumb, forefinger AND index finger.. you can achieve this.. though not sure it is very practical for shredding as I tend to hold the pick tighter when I am holding it like this.. this is the original way I used to hold my pick until I learnt it was easier to hold with just thumb and forefinger

This is where being able to switch from economy picking to alternate picking comes into play - being able to switch from up and down strokes to accomodate pinch harmonics etc. Personally I would never use or attempt to perfect ph's on upward strokes, it's far too awkward and likely to upset your rhythm.
The other alternative worth looking at is starting the riff/lick on the upstroke so that you when you get to the ph you can play it on a downstroke. It just comes about from lots of practice and being able to play fluidly. Eventually you just switch to hit a pinch harmonic without really thinking about it -mostly its a mix of alternate and economy picking combined with changing from upstroke to downstroke to fit the meter. All depends on what is most comfortable for the riff/lick and what comes after it.

Yeah... I spent a while practicing pinch harmonics on an upstroke... but it isn't usable in real situations in my opinion... I can't get the attack into the string on an upstroke.

I'd go with SH's suggestion of starting the lick on an upstroke so the harmonic lands on a down...

You clould try turning your fingers so that the pick is more perpendicular to the strings. This is how I play the intro to one of BLS's songs, I forget what song, I think its off the Blessed Hellride album.
