Before we start digging into all the great guitar parts in this song, let's briefly talk about the gear and the tones I'll be using for the different parts in this song, but as always it's really important to remember that you can play these parts on any guitar.
For the acoustic rhythm guitar I'm using my Martin-style VKV acoustic, and I'm getting the sound that you're hearing from a combination of my built-in pickup and the overhead mic that I'm also talking through.
For the lead guitar I'm using my Les Paul here on the bridge pickup, and I'm running that through a modeled version of a Fender amp with the volume cranked so I get some natural overdrive. There's very little bass in his tone- it's mostly just treble and midrange. So try to turn down the bass knob on your amp or a pedal if you have one. I also have a little bit of reverb and some fast, but subtle tremolo on it. But of course you can play this riff with any tone. Then for the lead guitar, I'm using almost the same tone, except I've lowered the amp volume a little bit so it's cleaner and cranked up the reverb quite a bit.
