Im confused about tuning.

What I am confused about is, does the fifth fret of the 6th string have to sound 100% equivalent to the open A string in standard tuning, and does the 5th fret of the A string have to sound 100% equivalent to the open D string...and so on.

That question is also applied to drop d tuning, would the 7th fret of the 6th string have to sound 100% equivalent to the open A string... and so on with the tuning of the other strings.

Help would be appreciated.

They will never sound EXACTLY the same... but similarity of the PITCH should be somethinglike 99.99% ...

If you're having problems like the guitar is perfectly tuned in the open position, and goes out of tune as you go up the neck, you should check :

1-The Intonation.

2-When was the last time you changed the strings?

What I wanted to know was if I played the 5th fret of the 6th string would it be the same as if I were to play the open A string?

I might try tuning with harmonics, I've heard that it's more accurate.

Yes, they should be te same...
Tuning with harmonics is more accurate since it makes you hear the 2 notes ringing together at the same time.

But if accuracy is what you're looking for , get yourself a tuner.

what i always do is use my Quiktune to tune it. (i'm just naming MY tuner, any tuner is fine of course)I make it fairly close according to my tuner. then i use harmonics to fine tune it quickly. this is probably the most effective method for tuning your guitar.
