I've been having trouble getting a good hard rock solo tone. My distortion/overdrive is nice, but when I play single notes it sounds terrible. I play through a Marshall AVT50 head with the matching 4x12 cab. Any suggestions/settings/effects to help? Thanks!

Increase your mids settings for the leads, also add enough reverbs and delay if available.

Some people like mids some people dont... i think mids probably do help.
Some delay and reverb should definitly make your tone sound better for solo's if you set it right !
I've just added some delay and reverb and it sounds pretty nice now :)
By virtue of their electrical properties, tubes generate a special waveform when they're saturated, which is why tube engineering has tremendous tonal advantages over solid state or DSP solutions, particularly for crunch and lead sounds. Tubes enter the saturation zone gradually or softly, which lends tube-driven tone its trademark yet totally unique character.

When I first started playing guitar , I was obsessed with mids scooping ... I'd usualy set the mids near 0 for heavy rhythms which sounded cool and use the same distortion for leads , and I didn't know why it sucked until I learned somewhere that mids are cool for lead tones.
Even guys with the heaviest rhythm distortions like Dimebag let their amps breathe some mids for leads.
