A locking tremolo has clamps that prevent the strings from slipping at the bridge saddles. This is supposed to prevent the strings from going out of tune when you do a mess of dive-bombing freaky stuff. It is usually paired with a locking nut which has clamps to keep the strings from going out of tune at that end. Of course, the strings go out of tune anyway, if they are not installed properly, and they stretch.
