It depends on your skill level, the difficulty of the song & the complexity of the tuning. For many songs with drop D, it's pretty obvious if you have any experience with that tuning & the ability to figure out songs in the first place.

On the other hand, sometimes I don't realize a song has non-standard tuning until I get to a certain chord voicing & realize it requires a certain tuning to get that particular voicing. That happens with Stones songs in which they occasionally have a guitar part in open G tuning for example.

It's easier to hear on songs with guitar parts that obviously have open strings that ring a lot & the note isn't one of the usual open strings.
