Fret out of tune

Question: my lower E string tunes perfectly. But when I hold down the second fret of the same string it plays a G instead of an F#. All of the other fretted notes sound correct. Should I still change out the string or could there be something else I'm missing?

Guitar setup isn't correct. Either the neck has bent, or intonation (free length of a string) is wrong. Usually wrong intonation doesn't affect that much as you described on lower frets. My bet is neck.
