Great question! The capo doesn't affect the tuning of the guitar, it will just change the pitch of the open strings and block access to any frets below it. So you should be able to play any leads from Guitar 2 without any issue.

So in this case, with the capo on the 3rd fret, the only thing to watch out for is that your open strings will be different (G C F Bb D G instead of E A D G B E) and you won't be able to fret anything on the 1st and 2nd strings.
