can you damage a 100 watt amp by playing it constantly at low volumes?

Playing at low volumes won't damage an amplifier. Higher volumes are what you've got to worry about. Even then, the chances of blowing a speaker are pretty slim.

So the tubes in a 100 watt amp played at low volumes are safe?

Absolutely - is there a reason you think it wouldn't be safe?
