Yeah, it depends on the pickups. If you have standard single coil pickups. those positions actually noise cancel (humbuck). You'll notice that any hum will go away in those positions. The sounds is kind of nasally and distinctive.

Originally I didn't use them either, until I realized Clapton, SRV, and Rory Gallagher would use those positions. Add a bit of gain on the amp or through something like a tube screamer and you can get some interesting tones.
