Do you mean from scratch or using a kit?
There's a company called PAiA that makes synth kits among other other things. I was going to build one of my own years ago (back in my mid teens), just to get the experience of working with the different modules and how they worked. However my parents talked me out of it. Kinda never forgave em cause the synth I wanted to get is a collectors item.

I got one of those huge module moog synths a few years back and enjoyed tweeking around with it.

I got the modules unasembled and learned a lot about how they work.
It's a crap load of work though and almost impossible to keep tuned.
Plus it's monophonic. It has an arpeggiator but there's no way to sync it with midi.
I only managed to get one song done and it took weeks to program just the bass parts.
