It shouldn't be a big deal. Depending on the tuners, the holes drilled into the headstock may need to be drilled to make the holes a bit bigger for the new tuners...but I'm sure they make locking tuners that will fit the stock holes.

Oh yeah, it will help a lot in keeping your strat in tune.
