It depends on where you get it.

Some places keep their guitars fairly well setup, others don't. If you buy your guitar from Musician's Friend or some other online place, you'll almost certainly have to take it to a local shop and have them do a setup.

When you settle on your guitar, you should be able to feel most of the things that would require adjustment -- either the strings being too damn high or so low that they're buzzing when you fret in certain places on the neck.

The other big thing is intonation which you'll be able to tell needs fixing if you tune the open strings and your chords don't sound right or if notes get more and more out of tune as you fret further and further up the neck. If you have an electric tuner, this is very easy to check.

If your guitarshop strings their guitars with really light strings and you play heavier ones, you're going to need to have at least that fixed.
