Try holding the plectrum so that only about 2mm protrudes for picking strings with. This may seem strange at the beginning but you will learn to give the plec different length by rolling your finger and thumb after a bit of practise.
To get a pinch harmonic, you must play on the down pick. As the plec crosses the string, the side of your thumb brushes the string. Hard to explain without having a visual. Just keep tryng it on the same note for a while, pick/brush then vibrato. Do it a hundred times untill you get it then practise on every fret on all six strings. Oh yeah, make sure you have a good distorted sound.

A smaller pick ,like a Dunlop Jazz III works great for pinch Harmonics. The smaller size puts your thumb closer to the strings, you may even hit some by accident!! Personally, I find a thicker pick helps too,and the JazzIII's are around 1.35 mm thick. Also, heavier strings can help. I use an 0.11 set and the added mass seems to make harmonics sound bigger. Zakk Wylde uses a 0.60 on his low "E" for christsakes!! He gets harmonics like crazy. On the other hand, Billy Gibbons of ZZ Top is the king of pinch harmonics and he says he uses an 0.008 set. I've got four Tele's and a Strat, the best guitar I have for squeals is my 87 MIJ toploader Tele. I dunno if it's the Basswood body, the 0.11-0.49 strings,or what. But that guitar squeals the better than the others.
Once you get it down, move your pick along the string to get other harmonics. You can change the pitch of the harmonic by moving either farther away from,or closer to the bridge.

I pinch the string between the fret and the end. It works every time.. Just do it in the exact center. I use a crappy pick and it still works. i've been experminenting with this for a couple of days now.. and i'm getting more used to it.

I've been training on creating nice pinch harmonics and this is what I found usefull:
- as superhuman says, keep that 2mm, nothing less because you will get pinch harmonics by accident, and nothing more because it's more difficult to brush the string with your thumb
- when you're accustomed with holding your plec like that, you hit the string and immediately after your plec hits the string, you turn your wrist a little bit outside making your thumb brush the string slightly making a pinch harmonic
- not all places sound just as good, so try to find your 'hotspots' on your guitar where you can get a nice pinch harmonic, the places are the same on each string
- to make the pinch harmonic really squeel you use vibrato a lot or simply bend it, also using your whammy bar can sometimes make one hell of a sound
- the more distortion/overdrive, the better you'll hear your pinch harmonic
- each guitar gives a different sound, so if you can practise on other guitars please do
- practise a lot! being able to make a pinch harmonic is one thing, but being able to do one during a song is way more difficult
- good luck! :)
= good music is good drinking =
