I think you're talking about a pinch harmonic. To do a pinch harmonic you crank up the gain, fret a note, then pick the note and touch the string lightly with you thumb as soon as you pick the note. That isn't a very good explanation but it is the best I can do without showing you...someone else might be able to tell you better. Oh and it helps if you bend the note as you pick it.
