A glass or metal slide will afford you more sustain. Or even ceramic. I got a Big Heart slide for about $10 at musician's friend, it's too heavy for my guitars, but it sounds excellent on bass (for Morphine type stuff).
As for the technique...I use standard tuning, and started out by trying to learn my lead licks on slide. It takes a bit of practice, but sounds soooo cool.
