I know this is going to sound kind of direct but...have you been through the Fundamentals courses yet? Those courses set the basis for what you're talking about.

If I understand correctly, you have a melody that you would play on one guitar and would like a harmony of that on another guitar. You first want to understand scales to know what note belong where in the various keys.

Once you have that, it is much easier to visualize on the fretboard what you need to do.

I would say looking through F1 and F2 is a must. After that, perhaps other instructors might have more specific advice.
