Ok this means that you play 1st 2nd 3rd then 4th fret on the low E String first. Then you play the same on the A, then the D, G, B and E. The whole thing is upside down because thats how you see it when the guitar is in your hands. So, the numbers indicate the fret to be played.

E--2- When they are all of in line like this, they are all to be played at the same time, so you play 4 on the D and A strings and 2 on the E. Its a powerchord.
